Try it after you loaded a LaTeX file.

Is d:/localtexmf configured as $TEXMLOCAL?  AUCTeX calls kpsewhich to
figure this out.  You can check it by executing
kpsewhich --progname latex --expand-path "$TEXMFLOCAL"
in a Windows prompt.
